Description

This sandwich ELISA kit from Antibodies.com enables the in vitro quantitative determination of human Lactate Dehydrogenase D (LDHD/DLD) in tissue homogenates, cell lysates, and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, coated with an immobilised antibody specific for LDHD/DLD. Standards and samples are added to the wells, followed by incubation with a Biotinylated Anti-LDHD/DLD Antibody and an HRP-Avidin conjugate. A TMB substrate solution is then added; catalysis produces a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and the concentration of LDHD/DLD is calculated by reference to a standard curve.

Quantitative performance is reported as an assay range of 1.56–100 ng/ml with a sensitivity of <0.59 ng/ml, and the assay can be completed in 4h 30m. The kit contains a pre-coated microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and 5 adhesive plate sealers. Components are stored at +4°C or -20°C as indicated; the kit should not be used past its expiration date.

Spike-and-recovery testing (n=5 per matrix) yielded average recoveries of 91% (range 80%–102%) in serum, 90% (81%–100%) in EDTA plasma, and 84% (80%–89%) in heparin plasma. Dilution linearity was evaluated at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma samples.
