Human Lactate Dehydrogenase C ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) enables in vitro quantitative determination of human Lactate Dehydrogenase C (LDH-C) in serum, plasma, tissue homogenates and other biological fluids. The kit uses a 96-well microtiter plate pre-coated with an antibody specific for LDH-C. Standards and test samples are added to the wells and LDH-C is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-LDH-C Antibody binds the captured target, followed by an HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, with colour intensity proportional to LDH-C concentration. Absorbance is read at 450 nm and quantified against a standard curve.

The assay is performed on a pre-coated microplate platform (12 x 8 well strips) with colourimetric detection and a total run time of 4h 30m. The reported assay range is 1.56–100 ng/ml with a sensitivity of 0.54 ng/ml; kit components are stored at +4°C or -20°C as instructed. Validation recovery for undiluted samples: serum (n=5) 80–102% (average 91%), EDTA plasma (n=5) 81–100% (average 90%), heparin plasma (n=5) 80–89% (average 84%). Dilution linearity from 1:2 to 1:16 is reported as serum 87–91% to 92–97%, EDTA plasma 90–105% to 79–108%, and heparin plasma 84–95% to 89–91%.
