Human Ki-67 Protein ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Ki67 protein in tissue homogenates, cell lysates, cell culture supernates, and other biological fluids.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for Ki67 pre-coated onto the plate. Standards and samples are added to the wells, where Ki67 is bound by the immobilized capture antibody. After washing, a biotinylated anti-Ki67 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the Ki67 concentration is calculated by referring to the standard curve.

The kit has a measuring range of 0.312-20 ng/ml and a sensitivity of 0.117 ng/ml. Total assay time is approximately 4h 30m. Reported recovery is 83%-100% in serum (average 91%, n=5), 91%-100% in EDTA plasma (average 95%, n=5), and 91%-101% in heparin plasma (average 96%, n=5). Kit components include the pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, to be stored at +4°C or -20°C as instructed.
