Human Keratin 8 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Cytokeratin 8 (Keratin 8) in serum, plasma, tissue homogenates, cell lysates, cell culture supernatants or other biological fluids. The kit is reactive with human samples and runs on a pre-coated microplate platform with a colorimetric readout read at 450 nm.

Standards and unknowns are captured by an immobilized antibody specific for Cytokeratin 8, followed by incubation with a biotinylated anti-Cytokeratin 8 antibody and an HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow after the acidic stop solution is added; absorbance is proportional to Cytokeratin 8 concentration and is read against a standard curve. The assay range is 0.156–10 ng/ml with a sensitivity of 0.057 ng/ml, and the total assay time is 4h 30m. Kit components are stored at +4 °C or −20 °C as instructed.

Performance characteristics reported in the product documentation include sample recovery (n=5) of 80–102% (average 91%) in serum, 81–100% (average 90%) in EDTA plasma and 80–89% (average 84%) in heparin plasma. Linearity across 1:2 to 1:16 dilutions ranged from 74% to 107% in serum, 79% to 108% in EDTA plasma and 82% to 105% in heparin plasma. The product is for research use only.
