Human Keratin 20 ELISA Kit

From Antibodies.com, this is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of human Cytokeratin 20 (also known as Keratin 20) in serum, plasma, tissue homogenates, or other biological fluids. The 96-well microplate is pre-coated with a Cytokeratin 20-specific capture antibody. Standards and test samples are pipetted into the wells and the Cytokeratin 20 present binds to the immobilised antibody. After washing, a biotinylated anti-Cytokeratin 20 detection antibody is added, followed by an HRP conjugate and TMB substrate. The HRP reaction produces a blue product which turns yellow upon addition of the acidic stop solution; the intensity of yellow, read at 450 nm on a colorimetric microplate reader, is directly proportional to the amount of Cytokeratin 20 captured in each well, and the concentration is calculated against the standard curve.

The kit is supplied on a pre-coated microplate comprising 12 x 8-well strips, with a quoted total assay time of 4h 30m. Quoted performance characteristics include a sensitivity of 0.064 ng/ml and a standard curve range of 0.156-10 ng/ml. In spike-and-recovery testing (n=5), recoveries were 83%-100% (average 91%) in serum, 90%-103% (average 96%) in EDTA plasma, and 90%-99% (average 94%) in heparin plasma. Linearity was evaluated across 1:2, 1:4, 1:8, and 1:16 dilutions in each matrix. Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
