Human Keratin 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com intended for the in vitro quantitative determination of human Cytokeratin 2e in serum, plasma, tissue homogenates and other biological fluids. The assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with results read colorimetrically at 450 nm. Reported sensitivity is 0.059 ng/ml and the working assay range is 0.156-10 ng/ml, with a total assay time of approximately 4 hours 30 minutes.

The procedure employs a biotinylated anti-Cytokeratin 2e detection antibody, an HRP-avidin conjugate and TMB substrate; the enzymatic reaction is stopped with an acidic stop solution to give a yellow product whose absorbance is proportional to the amount of analyte captured in each well. Supplied reagents include a lyophilized standard, detection solutions A and B, a 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, stored at +4°C or -20°C as instructed on the component label.

Spike-and-recovery testing (n=5) reported ranges of 80%-102% in serum (average 91%), 81%-100% in EDTA plasma (90%) and 80%-89% in heparin plasma (84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was demonstrated across serum, EDTA plasma and heparin plasma.
