Human Keratin 19 ELISA Kit

Supplied by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Cytokeratin 19 (Keratin 19) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs an antibody specific for Cytokeratin 19 pre-coated onto a 96-well microplate. Standards and samples are added to the wells and captured Cytokeratin 19 is detected using a Biotinylated Anti-Cytokeratin 19 Antibody, followed by an HRP-Avidin conjugate and TMB substrate. The resulting enzymatic product yields a yellow colour upon addition of stop solution, and the absorbance is read at 450nm in a microplate reader, with concentration calculated against a standard curve.

The stated product range is 12.5-800 pg/ml with a sensitivity of 5.4 pg/ml, and the complete assay time is approximately 4 hours 30 minutes. Sample recovery was evaluated in serum (average 91%, range 83-100%, n=5), EDTA plasma (average 96%, range 90-103%, n=5) and heparin plasma (average 95%, range 89-102%, n=5), with dilution linearity assessed at 1:2, 1:4, 1:8 and 1:16 across the same sample types. The kit is suitable for serum, plasma, cell lysates, tissue homogenates and cell culture supernates.

Components supplied include a pre-coated 96-well microplate (12 x 8 well strips), lyophilized standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
