Human Keratin 1 ELISA Kit

From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of human Cytokeratin 1 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. An antibody specific for Cytokeratin 1 has been pre-coated onto a 96-well microtiter plate arranged as 12 x 8 well strips.

The assay follows a sandwich enzyme immunoassay workflow. Standards and samples are added to the coated wells, captured Cytokeratin 1 is detected with a Biotinylated Anti-Cytokeratin 1 Antibody, and an HRP-Avidin conjugate catalyses TMB substrate to produce a colour change that is read at 450 nm in a colorimetric microplate reader. Cytokeratin 1 concentration is calculated by referring absorbance to the standard curve. The stated product range is 0.156-10 ng/ml with a sensitivity of 0.061 ng/ml, and the total assay time is 4h 30m.

Recovery has been reported in three matrices: serum (n=5) averaged 91% over a range of 80-102%, EDTA plasma (n=5) averaged 90% over 81-100%, and heparin plasma (n=5) averaged 84% over 80-89%. Linearity has been demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. Kit components include a pre-coated 96-well microplate, lyophilized standard (2 vials), Detection Solution A (120μl), Detection Solution B (120μl), 30X Wash Buffer (20ml), Sample Dilution Buffer (45ml), TMB Substrate (9ml), Stop Solution (6ml) and 5 adhesive plate sealers. Components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
