Human Karyopherin Alpha 2 ELISA Kit

The Antibodies.com Human Karyopherin Alpha 2 (KPNA2) ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of human KPNA2 in tissue homogenates, cell lysates, or other biological fluids. It is reactive with human samples and the assay is read on a colorimetric microplate reader.

The assay uses a microplate pre-coated with an antibody specific for KPNA2. Standards and samples are added and KPNA2 is captured by the immobilized antibody. After washing, a Biotinylated Anti-KPNA2 Antibody binds the captured KPNA2, followed by incubation with an HRP-Avidin conjugate. TMB substrate is then added to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of KPNA2 captured, and the concentration is calculated from O.D. absorbance at 450 nm referenced to a standard curve. Total assay time is 4h 30m.

The kit has a sensitivity of <57 pg/ml and an assay range of 125–8000 pg/ml. Components are stored at +4°C or -20°C as instructed. Reported recovery averages are 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5), and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity across 1:2 to 1:16 was demonstrated for serum, EDTA plasma, and heparin plasma.
