Human Kallikrein 9 ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human Kallikrein 9 in serum, plasma, tissue homogenates, and other biological fluids. It is intended for use with human samples and is read on a colorimetric microplate reader.

The kit employs the sandwich enzyme immunoassay technique using a 96-well microplate pre-coated with an antibody specific for Kallikrein 9. Standards and test samples are added to the wells, allowing the target to bind to the immobilized antibody. After washing, a biotinylated anti-Kallikrein 9 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then applied; HRP catalysis produces a blue colour that changes to yellow upon addition of the stop solution. Absorbance is read at 450 nm and the concentration of Kallikrein 9 in each sample is calculated by reference to the standard curve. The total assay time is 4 hours 30 minutes.

Quoted performance figures from the manufacturer include a sensitivity of <0.055 ng/ml and an assay range of 0.156–10 ng/ml. Reported recovery ranges (n=5) are 80–102% for serum, 81–100% for EDTA plasma, and 80–89% for heparin plasma. Dilution linearity has been assessed at 1:2, 1:4, 1:8, and 1:16 dilutions, with ranges of 87–91%, 87–107%, 74–101%, and 92–97% respectively for serum; 90–105%, 84–101%, 90–101%, and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% for heparin plasma. The kit contains a pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with components stored at +4°C or -20°C as indicated on the product insert.
