Human Kallikrein 5 ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human Kallikrein 5 in serum, plasma, saliva, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate configured as 12 x 8 well strips and is read on a colorimetric microplate reader at 450 nm. It has a stated assay range of 78.1-5000 pg/ml with a sensitivity of 29 pg/ml, and the full protocol runs in approximately 4 hours 30 minutes.

The workflow follows a standard sandwich immunoassay format. An antibody specific for Kallikrein 5 is immobilised on the plate; standards and samples are added, then the wells are washed and incubated with a biotinylated anti-Kallikrein 5 antibody. HRP-Avidin conjugate is added, and TMB substrate is used to visualise the enzymatic reaction as a blue product that turns yellow after the acidic stop solution is added. The density of yellow is proportional to the amount of Kallikrein 5 captured in each well, with concentration calculated by reading the absorbance at 450 nm and referring to the standard curve.

Reported recovery ranges include 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma, and 80%-89% (average 84%) for heparin plasma across dilution series of 1:2, 1:4, 1:8 and 1:16.
