Human Kallikrein 3 ELISA Kit

A sandwich ELISA kit from Antibodies.com for the in vitro quantitative determination of human Kallikrein 3/KLK3. The assay is compatible with serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Quantitative readout is colorimetric, obtained by measuring the absorbance at 450 nm on a standard microplate reader after the TMB substrate has been converted from blue to yellow by addition of acidic stop solution.

The kit covers a measuring range of 1.95–125 ng/ml with a sensitivity of <0.7 ng/ml, and the full protocol can be completed in 4h 30m. Components should be stored at +4 °C or –20 °C as indicated, and the kit must not be used past its expiration date. Spike-and-recovery testing in human samples (n=5 per matrix) returned average recoveries of 91% in serum (range 80%–102%), 90% in EDTA plasma (range 81%–100%) and 84% in heparin plasma (range 80%–89%).

The sandwich format uses an antibody specific for Kallikrein 3/KLK3 immobilised on the plate, a biotinylated anti-KLK3 detection antibody and an HRP–avidin conjugate. Sample dilutions tested at 1:2, 1:4, 1:8 and 1:16 gave recovery ranges of 87–91%, 87–107%, 74–101% and 92–97% in serum; 90–105%, 84–101%, 90–101% and 79–108% in EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% in heparin plasma.
