Description

This sandwich Enzyme-Linked Immunosorbent Assay, manufactured for Antibodies.com, is intended for the in vitro quantitative determination of human Kallikrein 10/K10 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs a pre-coated 96-well microplate to which an antibody specific for Kallikrein 10/K10 has been immobilised. Standards and test samples are added to the wells and the Kallikrein 10/K10 present is captured by the immobilised antibody. After washing, a Biotinylated Anti-Kallikrein 10/K10 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Kallikrein 10/K10 captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to a standard curve.

Reactivity is human, and the kit is supplied on a pre-coated microplate platform of 12 × 8-well strips. The quantitative measurement range is 0.156–10 ng/ml with a sensitivity of <0.048 ng/ml, and the assay takes approximately 4 hours 30 minutes to perform. Reported average recovery values from spiked samples (n=5) are 91% in serum (range 83–100%), 96% in EDTA plasma (range 90–103%) and 95% in heparin plasma (range 89–102%). Linearity of dilution has been demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions in serum, EDTA plasma and heparin plasma. Kit components are intended to be stored at +4 °C or –20 °C as instructed, and the kit should not be used past its expiration date.
