This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Junctional Adhesion Molecule 2/JAM-B in serum, plasma, tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, in which an antibody specific for Junctional Adhesion Molecule 2/JAM-B is pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, and any target present is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-Junctional Adhesion Molecule 2/JAM-B Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of JAM-B captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.
The kit has a working range of 31.2–2000 pg/ml and a sensitivity of <12.6 pg/ml. It is intended for use with a colorimetric microplate reader, with a total assay time of 4h 30m. The pre-coated plate is supplied as 12 x 8 well strips. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
The manufacturer documentation reports recovery and dilution linearity data for serum, EDTA plasma and heparin plasma. Average recovery is reported as 91% from serum (range 80–102%, n=5), 90% from EDTA plasma (range 81–100%, n=5) and 84% from heparin plasma (range 80–89%, n=5). Dilution linearity at sample dilutions of 1:2, 1:4, 1:8 and 1:16 is also provided across the same three sample types, with reported ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5).
