Description
A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Integrin alpha 5 (also known as CD49e, FNRA, VLA-5 and ITGA5) in serum, plasma, tissue homogenates, and other biological fluids. The assay employs a pre-coated microplate with an immobilised capture antibody specific for Integrin alpha 5. Standards and samples are added to the wells, where the target binds to the capture antibody. Following incubation and washing, a biotinylated detection antibody is added to form a sandwich complex, which is visualised using HRP-Streptavidin conjugate and TMB substrate. The enzymatic reaction yields a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm and concentration is calculated against the standard curve.
The antibody pair is specific for Integrin alpha 5, with no reported cross-reactivity with other analogs or variants. Detection is colorimetric and the total assay time is 4 hours. Validated recovery ranges are 88–102% (serum, n=10, average 93%), 88–99% (EDTA plasma, n=10, average 94%) and 86–101% (heparin plasma, n=10, average 98%). Serum and plasma samples should be diluted at least 1:2 with Sample Dilution Buffer prior to testing. The kit is shipped at +4°C and components are stored at +4°C or −20°C according to the protocol manual; freeze/thaw cycles should be avoided and the kit must not be used past its expiration date.
