The assay employs a pre-coated microplate in a 12 x 8 well strip format with an antibody specific for Involucrin immobilised on the wells. Standards and samples are added, followed by a Biotinylated Anti-Involucrin Antibody and an HRP-Avidin conjugate for detection. A TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of Involucrin captured and is read at 450nm in a colorimetric microplate reader. The product range is 0.312–20 ng/ml with a sensitivity of <0.116 ng/ml, and the total assay time is 4h 30m.

Reported sample recovery (n=5) averages 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%). Dilution linearity is documented at 1:2, 1:4, 1:8 and 1:16 dilutions across the same sample types. Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
