The Antibodies.com sandwich ELISA kit enables the in vitro quantitative determination of human CXCR2 (Interleukin 8 Receptor Beta) in tissue homogenates, cell lysates and other biological fluids. A 96-well microtiter plate is pre-coated with an antibody specific for CXCR2, and standards or samples are added so that CXCR2 in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-CXCR2 Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm using a colorimetric microplate reader, and the concentration of CXCR2 is calculated from the standard curve.

The assay has a sensitivity of 0.25 ng/ml and a standard range of 0.781–50 ng/ml, with a total assay time of 4 hours 30 minutes. Reported sample recovery values include serum (n=5) ranging from 80% to 102% with an average of 91%, EDTA plasma (n=5) ranging from 81% to 100% averaging 90%, and heparin plasma (n=5) ranging from 80% to 89% averaging 84%. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
