Human Interleukin-8 ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Interleukin-8 (IL-8) in serum, plasma, and cell culture supernatant. The assay uses a quantitative sandwich immunoassay format in which standards or samples are incubated in microtitre wells pre-coated with a monoclonal antibody specific for IL-8, followed by a standardised horseradish peroxidase (HRP)-conjugated monoclonal antibody and a TMB (3,3',5,5'-tetramethyl-benzidine) substrate solution, with absorbance read at 450 nm ± 2 nm after the reaction is terminated by the addition of a sulphuric acid solution.

Performance characteristics include an assay range of 10-1600 pg/ml and a sensitivity of < 10 pg/ml. The kit shows no detectable cross-reactivity with human monocyte chemotactic activating factor (MCAF) or RANTES, and recognises both natural and recombinant human IL-8.

The colourimetric assay has a total run time of approximately 2h 30m, and the kit is shipped and stored at 4°C. Two calibrator diluents are supplied: Calibrator Diluent I for serum/plasma testing and Calibrator Diluent II for cell culture supernatant and urine testing. Sample IL-8 concentrations are calculated by comparing sample optical density to a standard curve generated by two-fold serial dilution of the provided standard with the appropriate calibrator diluent.
