Human Interleukin 7 Receptor ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human CD127 (Interleukin 7 Receptor) in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for CD127 has been immobilised. Standards and test samples are added to the wells, the plate is incubated with a Biotinylated Anti-CD127 Antibody, and an HRP-Avidin conjugate is then applied. A TMB substrate solution is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of the acidic stop solution; the density of yellow is proportional to the amount of CD127 captured in each well.

The kit has a stated sensitivity of <0.64 ng/ml and a working assay range of 1.56–100 ng/ml, with a total assay time of 4h 30m. Reported recovery values are: serum 83–100% (average 91%, n=5), EDTA plasma 91–100% (average 95%, n=5) and heparin plasma 91–101% (average 96%, n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is also reported across these sample types. Kit components include lyophilised standard (2 vials), detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored at +4°C or −20°C as indicated on the component table. Absorbance is read at 450 nm in a colorimetric microplate reader and the CD127 concentration is calculated against the standard curve.
