Description

Manufactured by Antibodies.com and categorised under Assays and Kits, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Interleukin-6 in serum, plasma, cell culture supernatant, and other biological fluids. The assay applies a quantitative sandwich immunoassay technique using a microplate pre-coated with a monoclonal antibody specific to IL-6. Standards or samples are added together with a biotin-conjugated polyclonal antibody preparation specific for IL-6, and any IL-6 present becomes immobilised and sandwiched between the coating antibody and the biotin conjugate.

Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well, and a TMB (3,3',5,5'-tetramethyl-benzidine) substrate solution is then introduced. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the resulting colour change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The kit recognises both natural and recombinant human IL-6 and has shown no cross-reactivity with various other cytokines superfamily proteins. Reported sensitivity is < 7 pg/ml, with a product range of 7–2000 pg/ml. The pre-coated microplate platform comprises 12 x 8 well strips, the detection method is colorimetric on a microplate reader, and the total assay time is 3 hours 30 minutes. The kit is shipped and stored at 4 °C and includes two calibration diluents: Calibrator Diluent I for serum/plasma testing and Calibrator Diluent II for cell culture supernatant testing. The provided standard is diluted 2-fold with the appropriate Calibrator Diluent and assayed alongside samples to generate a standard curve of Optical Density (O.D.) versus IL-6 concentration (pg/mL).
