Detection is colourimetric, with the enzymatic reaction visualised using TMB substrate and quantified by measuring absorbance at 450nm on a microplate reader.

The reported sensitivity is <5.8 pg/ml, with a quantitative assay range of 15.6-1000 pg/ml and a total assay time of 4h 30m. Spike-and-recovery testing yielded average recoveries of 91% in serum (n=5, range 83-100%), 94% in EDTA plasma (n=5, range 91-98%), and 94% in heparin plasma (n=5, range 90-99%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across the same sample types.
