Human Interleukin 35 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human EBI3, associated with Interleukin 35, in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for EBI3, with a quoted detection range of 15.6–1000 pg/ml and a sensitivity of 5.6 pg/ml. Standards and samples are incubated on the plate and washed, followed by incubation with a biotinylated anti-EBI3 detection antibody, addition of an HRP-avidin conjugate and visualisation with TMB substrate, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the EBI3 concentration is calculated against the standard curve.

The full assay time is 4 hours 30 minutes, and the kit is intended for research use only. Kit components are stored at +4°C or –20°C as instructed and should not be used past the expiration date. In validation studies the average recovery was 91% in serum (range 83–100%, n=5), 94% in EDTA plasma (range 91–98%, n=5) and 94% in heparin plasma (range 90–99%, n=5). Supplied reagents include the pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard, Detection Solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers.
