Human Interleukin 26 ELISA Kit

The Antibodies.com sandwich Enzyme-Linked Immunosorbent Assay is designed for the in vitro quantitative determination of human IL-26 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique: an antibody specific for IL-26 is pre-coated onto a 96-well microtiter plate, standards and samples are added, and the captured IL-26 is detected with a Biotinylated Anti-IL-26 Antibody followed by HRP-Avidin and TMB substrate, producing a blue colour that turns yellow upon addition of stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration calculated against a standard curve.

The assay has a standard range of 15.6–1000 pg/ml and a sensitivity of 6.1 pg/ml. It runs on a pre-coated 96-well plate (12 x 8 well strips) with a total assay time of 4h 30m. Provided kit components include the pre-coated 96 well microplate stored at -20°C, 2 vials of lyophilized standard, Detection Solution A (120μl) and Detection Solution B (120µl) both at -20°C, Wash Buffer 30X (20ml) at +4°C, Sample Dilution Buffer (45ml) at -20°C, TMB Substrate (9ml) at +4°C, Stop Solution (6ml) at +4°C, and 5 adhesive plate sealers.

Reported recovery (n=5) averaged 91% for serum (range 83%–100%), 96% for EDTA plasma (range 90%–103%) and 94% for heparin plasma (range 90%–99%). Linearity across 1:2, 1:4, 1:8 and 1:16 dilutions (n=5) was 90–99%, 82–108%, 82–96% and 92–103% for serum; 84–105%, 88–94%, 89–107% and 72–106% for EDTA plasma; and 89–92%, 88–108%, 84–102% and 90–98% for heparin plasma.
