The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for IL-18R beta/IL-18RAP pre-coated onto a microplate. Standards and test samples are added to the wells, followed by a Biotinylated Anti-IL-18R beta/IL-18RAP Antibody and an HRP-Avidin conjugate. A TMB substrate produces a blue product that turns yellow upon addition of the acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader.

The kit reports an assay sensitivity of <0.051 ng/ml and a working range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Reported recovery values from sample testing include serum (range 80–102%, average 91%), EDTA plasma (range 81–101%, average 91%) and heparin plasma (range 80–89%, average 84%), with additional dilution linearity data reported for serum, EDTA plasma and heparin plasma across multiple dilutions.
