Human Interleukin 17D ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of human Interleukin 17D (IL-17D) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay is reactive with human samples and runs on a pre-coated microplate arranged as 12 x 8 well strips. An antibody specific for IL-17D is immobilised on the plate; standards and test samples are added and any IL-17D present is bound, followed by a Biotinylated Anti-IL-17D Antibody and an HRP-Avidin conjugate. TMB substrate solution is then added to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a colourimetric microplate reader and the concentration of IL-17D is calculated by reference to the standard curve.

The standard curve covers a product range of 15.6-1000 pg/ml, with a sensitivity of 5.3 pg/ml. In spike-and-recovery testing (n=5 per sample type), average recoveries were 91% for serum (range 83%-100%), 95% for EDTA plasma (range 91%-100%) and 96% for heparin plasma (range 91%-101%). Linearity was demonstrated across serial dilutions of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. The total assay time is 4h 30m.

Kit components include a pre-coated 96 well microplate, lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
