From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Interleukin-13 Receptor Alpha 2 in serum, plasma and cell culture supernatant. The assay applies the quantitative sandwich enzyme immunoassay technique, using a monoclonal antibody specific to IL-13 Ra2 pre-coated onto a microplate, a biotin-conjugated antibody preparation specific for IL-13 Ra2, and avidin-Horseradish Peroxidase (HRP) for colorimetric detection. The platform is a pre-coated microplate of 12 x 8 well strips.

Reported performance characteristics include a sensitivity of < 10 pg/ml and an assay range of 10-500 pg/ml. The kit has shown no cross-reactivity with other cytokines and growth factors tested, and the sandwich ELISA recognises both natural and recombinant human IL-13 Ra2. The reagent package includes two calibration diluents — Calibrator Diluent I for serum/plasma testing and Calibrator Diluent II for cell culture supernatant testing — and a provided standard that is diluted 2-fold with the appropriate Calibrator Diluent to produce a standard curve of Optical Density versus IL-13 Ra2 concentration. Reagents are shipped and stored at 4°C and the assay is intended for use with a colorimetric microplate reader.

IL-13 receptor alpha 2, also known as CD213α2, is a subunit of the IL-13 receptor complex encoded by the gene locus Xq13.1-q28 on chromosome X. It lacks a cytoplasmic domain and, although it binds IL-13 with high affinity, it does not appear directly involved in typical IL-13-stimulated signal transduction; its binding to IL-13 is IL-4R independent and is not associated with Jaks or the phosphorylation and activation of Stats. Soluble IL-13Rα2 may serve as a decoy receptor to dampen the cell reaction to IL-13 stimulation. IL-13Rα2 exists both on the cell membrane and in soluble form, and is relatively rich in hemopoietic or hypervascular tissues, while expressed at low levels or not expressed in other tissues. The gene was initially cloned from a kidney cell line and has also been found in brain, spleen, liver, thymus and bone marrow. Abnormal IL-13Rα2 expression has been associated with pathological conditions including susceptibility to Systemic Sclerosis, and overexpression has been found in brain tumors and ovarian cancer, making IL-13Rα2 a candidate biomarker and target for immunotherapy.
