Human Interleukin-10 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Interleukin-10 in serum, plasma, cell culture supernatant, and other biological fluids. The assay applies a quantitative sandwich immunoassay format using a pre-coated microplate (12 x 8 well strips) bearing a monoclonal antibody specific for IL-10.

Standards or samples are co-incubated with a biotin-conjugated monoclonal antibody specific for IL-10, followed by detection with Avidin conjugated to Horseradish Peroxidase (HRP) and a TMB (3,3′,5,5′-tetramethyl-benzidine) substrate solution. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the colour change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm using a colorimetric microplate reader.

The kit shows no cross-reactivity with other cytokines tested and detects both natural and recombinant human IL-10. Sensitivity is < 4 pg/ml and the assay range is 4–1000 pg/ml. Two calibration diluents are supplied: Calibrator Diluent I for serum/plasma testing and Calibrator Diluent II for cell culture supernatant testing, with the provided standard diluted 2-fold. The total assay time is 3h 30m and the kit is shipped and stored at 4°C.
