Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the quantitative in vitro determination of human Intercellular Adhesion Molecule 2 (ICAM2). The kit is reactive with human samples and is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids.

A microplate is pre-coated with an antibody specific for ICAM2. Standards and test samples are added to the wells and ICAM2 present in each sample is bound by the immobilized antibody. The wells are washed and then incubated with a biotinylated anti-ICAM2 antibody that binds the captured ICAM2. HRP-avidin conjugate is added next, followed by TMB substrate solution, which catalyses a blue-coloured product that changes to yellow after addition of the acidic stop solution. The intensity of yellow is proportional to the amount of ICAM2 captured, and the concentration is calculated by reading the absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

The reported product range is 6.25–400 ng/ml and the sensitivity is <1.6 ng/ml, with an assay time of 4h 30m and colorimetric detection. Recovery in serum (n=5) ranges from 80% to 102% (average 91%), in EDTA plasma (n=5) from 81% to 100% (average 90%), and in heparin plasma (n=5) from 80% to 89% (average 84%). Kit components should be stored at +4°C or -20°C as instructed in the protocol.
