Human Inter Alpha-Globulin Inhibitor H5 ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the quantitative in vitro determination of human ITIH5 (Inter Alpha-Globulin Inhibitor H5) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay has a product range of 78.1–5000 ng/ml and a sensitivity of 33 ng/ml, read on a colorimetric microplate reader at 450 nm using a pre-coated 96-well plate (12 x 8 well strips), with a total assay time of 4h 30m.

The procedure uses the sandwich enzyme immunoassay technique: an antibody specific for ITIH5 is pre-coated onto the plate, standards and test samples are added and bound, wells are washed, then Biotinylated Anti-ITIH5 Antibody and HRP-Avidin conjugate are added sequentially. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution; absorbance density is proportional to ITIH5 concentration.

Validation showed 80%–102% recovery for serum (average 91%), 81%–100% for EDTA plasma (average 90%), and 80%–89% for heparin plasma (average 84%), with linear dilution (1:2 to 1:16) performance ranging 74%–107% across serum, 79%–108% for EDTA plasma, and 82%–105% for heparin plasma. Kit components include pre-coated microplate strips, lyophilized standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, stored at +4°C or –20°C as instructed; do not use past expiration date.
