From Antibodies.com, this sandwich enzyme-linked immunosorbent assay (ELISA) is designed for the in vitro quantitative determination of human ITIH2/SHAP (Inter Alpha-Globulin Inhibitor H2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit uses an antibody specific for ITIH2/SHAP pre-coated onto a microplate, with captured analyte detected using a biotinylated anti-ITIH2/SHAP antibody, HRP-avidin conjugate, and TMB substrate, producing a colour change read at 450 nm on a colorimetric microplate reader.

The quantitative range spans 78.1-5000 pg/ml with a sensitivity of <35 pg/ml, and the assay is completed in 4h 30m. Kit components are stored at +4°C or -20°C as instructed. Reported spike-and-recovery values were 80%-102% (average 91%) in serum, 81%-100% (average 90%) in EDTA plasma, and 80%-89% (average 84%) in heparin plasma (n=5 for each sample type).

Dilution linearity across 1:2, 1:4, 1:8, and 1:16 sample dilutions (n=5 per sample type) was reported at 87-91%, 87-107%, 74-101%, and 92-97% for serum; 90-105%, 84-101%, 90-101%, and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105%, and 89-91% for heparin plasma.
