Human Insulin Like Growth Factor Binding Protein, Acid Labile Subunit ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com designed for the in vitro quantitative determination of human Insulin Like Growth Factor Binding Protein, Acid Labile Subunit (ALS) in serum, plasma, tissue homogenates or other biological fluids. The assay employs a sandwich enzyme immunoassay technique: an antibody specific for ALS is pre-coated onto a 96-well microplate, standards and test samples are added to the wells, and captured ALS is detected using a Biotinylated Anti-ALS Antibody followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the ALS concentration is calculated against a standard curve.

Reported assay range is 1.56–100 ng/ml with a sensitivity of 0.65 ng/ml. Total assay time is approximately 4 hours 30 minutes. The kit is supplied with a pre-coated 96 well microplate (12 x 8 well strips), lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored at +4°C or −20°C as instructed and not used past the expiration date.

Average recovery in neat samples (n=5) is reported as 91% for serum (range 80–102%), 91% for EDTA plasma (range 81–101%) and 84% for heparin plasma (range 80–89%). Dilution linearity has been characterised at 1:2, 1:4, 1:8 and 1:16 dilutions in serum, EDTA plasma and heparin plasma (n=5 for each matrix).
