The assay employs a pre-coated microplate formatted as 12 x 8 well strips, with an immobilised antibody specific for IKK beta. Standards and samples are added to the wells, followed by a biotinylated anti-IKK beta detection antibody and an HRP-avidin conjugate. The enzymatic reaction is visualised using TMB substrate, producing a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450nm in a microplate reader.
