A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human INHBC (Inhibin Beta C) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for INHBC pre-coated onto a microplate. Standards and test samples are added to the wells and INHBC present is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-INHBC Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution.

The density of yellow is proportional to the amount of INHBC captured in each well, and concentration is calculated from the O.D. absorbance read at 450nm in a microplate reader against a standard curve. The quantitative sandwich assay has a reported product range of 0.156–10 ng/ml and a sensitivity of <0.062 ng/ml. Total assay time is 4h 30m, with detection performed on a colorimetric microplate reader.

Manufacturer-reported recovery values include an average of 91% in serum (range 83%–99%, n=5), 89% in EDTA plasma (range 80%–99%, n=5), and 90% in heparin plasma (range 82%–99%, n=5). Linearity values are reported across 1:2, 1:4, 1:8 and 1:16 dilutions for each sample type. Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
