A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Inducible T-Cell Co-Stimulator (ICOS) in serum, plasma, urine, cell lysates and other biological fluids. The assay is reactive with human samples and is run on a colorimetric microplate reader using a pre-coated 96-well microplate arranged as 12 x 8 well strips.

The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for ICOS pre-coated onto the microplate wells. Standards and test samples are added and ICOS present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-ICOS Antibody is added, followed by HRP-Avidin conjugate and TMB substrate. The HRP enzymatic reaction produces a blue-coloured product that changes to yellow upon addition of acidic stop solution. The concentration of ICOS is calculated by reading the O.D. absorbance at 450nm and referring to the standard curve. The assay has a sensitivity of <6.2 pg/ml, a standard range of 15.6-1000 pg/ml, and a total assay time of 4 hours 30 minutes.

Reported validation values for serum (n=5) are a range of 80%-102% with an average of 91%; EDTA plasma (n=5) gives a range of 81%-100% with an average of 90%; and heparin plasma (n=5) gives a range of 80%-89% with an average of 84%. Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date.
