Human Iduronidase Alpha L ELISA Kit

The Antibodies.com Human IDUA ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) designed for the in vitro quantitative determination of human IDUA in tissue homogenates, cell lysates, or other biological fluids. The assay employs a pre-coated microplate in a 12 x 8 well strip format, with an antibody specific for IDUA immobilised on the plate. Standards and samples are added, followed by incubation with a Biotinylated Anti-IDUA Antibody and then an HRP-Avidin conjugate. Detection is colourmetric, using TMB substrate solution that produces a yellow product after addition of acidic stop solution, with the optical density read at 450 nm in a microplate reader.

The quoted assay range is 1.56–100 ng/ml and the stated sensitivity is <0.63 ng/ml. Total assay time is 4h 30m. Recovery has been demonstrated in serum (n=5, range 80%–102%, average 91%), EDTA plasma (n=5, range 81%–100%, average 90%) and heparin plasma (n=5, range 80%–89%, average 84%). Dilution linearity is reported across 1:2, 1:4, 1:8 and 1:16 sample dilutions for serum, EDTA plasma and heparin plasma. Kit components include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers, with storage at +4°C or −20°C as instructed.
