Description

Supplied under the Antibodies.com brand, this kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human ORP150 (Hypoxia Up Regulated 1) in serum, plasma, tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for ORP150 is pre-coated onto a 96-well microtiter plate formatted as 12 x 8 well strips. Standards and test samples are added to the wells and the ORP150 present is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-ORP150 Antibody is added, followed by an HRP-Avidin conjugate and further incubation. A TMB substrate solution is then used to visualise the HRP enzymatic reaction; the resulting yellow product (after addition of acidic stop solution) is read at 450 nm on a colorimetric microplate reader, and the ORP150 concentration is calculated by reference to the standard curve.

Reactivity: Human. Assay type: sandwich (quantitative). Detection: colorimetric. Total assay time: 4h 30m. Reported sensitivity: <0.061 ng/ml. Assay range: 0.156-10 ng/ml. Kit components are stored at +4°C or -20°C as instructed and must not be used past the stated expiration date.

Sample recovery (n=5) for ORP150 has been characterised as follows: serum 80-102% with an average of 91%; EDTA plasma 81-100% with an average of 90%; heparin plasma 80-89% with an average of 84%. Dilution linearity has also been examined across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma samples.
