From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human HIF-2 alpha in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay uses the sandwich enzyme immunoassay technique with a pre-coated microplate. An antibody specific for HIF-2 alpha is immobilised on the plate; standards and test samples are added and the target is captured. After washing, a Biotinylated Anti-HIF-2 alpha Antibody is added followed by HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the HIF-2 alpha concentration is calculated against the standard curve.

The product range is 0.156–10 ng/ml with a sensitivity of <0.053 ng/ml, and total assay time is 4 hours 30 minutes. Kit components should be stored at +4 °C or –20 °C as instructed and not used past the expiration date. Recovery averaged 91% in serum (range 83%–100%), 96% in EDTA plasma (90%–103%) and 94% in heparin plasma (90%–99%). Dilution linearity has been demonstrated in serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16.
