From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of human Hypoxia Inducible Factor 1 Alpha (HIF-1 alpha) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. A 96-well microplate is pre-coated with an antibody specific for HIF-1 alpha. Standards and samples are added to the wells; HIF-1 alpha is captured by the immobilized antibody. After washing, a biotinylated anti-HIF-1 alpha antibody and HRP-Avidin conjugate are added. TMB substrate produces a blue-coloured product that changes to yellow upon addition of acidic stop solution; absorbance is read at 450 nm and the concentration is determined from the standard curve.

Key specifications include a measuring range of 0.156-10 ng/ml, a sensitivity of 0.055 ng/ml, colorimetric detection, and an assay time of 4h 30m, read on a colorimetric microplate reader. Spike-and-recovery testing (n=5) showed average recoveries of 91% in serum, 96% in EDTA plasma, and 95% in heparin plasma. Dilution linearity was demonstrated across 1:2 to 1:16 in all three sample types.
