The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for HMGCS2 pre-coated onto a microplate. Standards and samples are added, followed by a Biotinylated Anti-HMGCS2 Antibody and HRP-Avidin conjugate, with a TMB substrate producing a yellow product whose absorbance at 450 nm is proportional to HMGCS2 concentration.

The kit reports a product range of 0.156-10 ng/ml and a sensitivity of <0.053 ng/ml. It is read on a colorimetric microplate reader, with a total assay time of 4h 30m, and kit components are stored at +4°C or -20°C. Manufacturer recovery data for serum (n=5) ranged 80%-102% with an average of 91%, for EDTA plasma (n=5) 81%-100% averaging 90%, and for heparin plasma (n=5) 80%-89% averaging 84%. Dilution linearity was reported across 1:2 to 1:16 dilutions for serum, EDTA plasma, and heparin plasma samples.
