Human Hydroxymethylbilane Synthase ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human HMBS/PBGD in tissue homogenates, cell lysates, or other biological fluids. The assay employs an antibody specific for HMBS/PBGD pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where target is captured by the immobilized antibody. After washing, a Biotinylated Anti-HMBS/PBGD Antibody binds the captured analyte, and an HRP-Avidin conjugate is then added. TMB substrate solution catalyses a blue-coloured product that changes to yellow upon addition of acidic stop solution; the density of yellow, read at 450nm on a colorimetric microplate reader, is proportional to the amount of HMBS/PBGD present in each well.

The kit is reactive with human samples and reports a quantitative assay range of 0.781-50 ng/ml with a sensitivity of 0.27 ng/ml. Total assay time is approximately 4h 30m. Manufacturer validation data report recoveries from spiked serum (80%-102%, average 91%; n=5), EDTA plasma (81%-100%, average 90%; n=5), and heparin plasma (80%-89%, average 84%; n=5). Dilution linearity data are provided for serum, EDTA plasma, and heparin plasma at 1:2, 1:4, 1:8, and 1:16 dilutions.

Kit components include a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers.
