Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Huntingtin in tissue homogenates, cell lysates, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, in which an antibody specific for Huntingtin is pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, where Huntingtin is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Huntingtin Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Huntingtin captured in each well, and concentration is calculated from the O.D. absorbance at 450 nm read in a microplate reader, by reference to a standard curve.

The assay has a sensitivity of <0.064 ng/ml and a product range of 0.156-10 ng/ml. Detection is colourmetric on a pre-coated microplate platform (12 x 8 well strips), with a total assay time of 4h 30m. It is reactive with human samples and is suitable for use with cell lysates and tissue homogenates. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Recovery data (n=5): serum 80-102% (average 91%), EDTA plasma 81-100% (average 90%), heparin plasma 80-89% (average 84%). Linearity data across dilutions of 1:2, 1:4, 1:8, and 1:16 (n=5): serum 74-107%, EDTA plasma 79-108%, heparin plasma 82-105%.
