Description
From Antibodies.com, this assay is designed for the in vitro quantitative determination of human HDAC9 (Histone Deacetylase 9) in tissue homogenates, cell lysates or other biological fluids. It employs a sandwich ELISA format with reactivity limited to human samples.
The kit uses a pre-coated microplate onto which an antibody specific for HDAC9 has been immobilised. Standards and test samples are added to the wells and any HDAC9 present is bound by the immobilised antibody. After washing, a biotinylated anti-HDAC9 detection antibody is added, followed by HRP-avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of HDAC9 captured in each well. Concentration is calculated by reading absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.
Reported performance characteristics include a sensitivity of <0.138 ng/ml, an assay range of 0.312–20 ng/ml, and a total assay time of 4h 30m. In spike-and-recovery testing, serum samples (n=5) showed a range of 80%–102% with an average of 91%, EDTA plasma (n=5) showed 81%–100% with an average of 90%, and heparin plasma (n=5) showed 80%–89% with an average of 84%. Kit components should be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
