Human Histone Deacetylase 6 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com for the in vitro quantitative determination of human Histone Deacetylase 6 (HDAC6) in tissue homogenates, cell lysates, or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for HDAC6. Standards and samples are added to the wells, where HDAC6 is captured by the immobilised antibody. After washing, a Biotinylated Anti-HDAC6 Antibody binds the captured analyte, followed by addition of HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader.

The kit reports an assay range of 0.312-20 ng/ml and a sensitivity of <0.114 ng/ml, with a total assay time of 4 hours 30 minutes. Detection is quantitative and colorimetric on a pre-coated 96-well plate configured as 12 x 8 well strips. Sample performance has been characterised in serum, EDTA plasma and heparin plasma, with mean recoveries of 91%, 90% and 84% respectively (n=5). Dilutional linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions, with recovery ranges of 74-107% in serum, 79-108% in EDTA plasma and 82-105% in heparin plasma across these dilutions.
