Human Histone Acetyltransferase 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human KAT1/HAT1 in tissue homogenates or other biological fluids. The assay is reactive with human samples and uses a pre-coated 96-well microplate.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for KAT1/HAT1 is pre-coated onto the plate; standards and test samples are added and the target binds to the immobilized antibody. After washing, a Biotinylated Anti-KAT1/HAT1 Antibody is added, followed by HRP-Avidin conjugate and TMB substrate solution. The enzymatic reaction produces a blue-coloured product that changes to yellow upon addition of acidic stop solution, with absorbance read at 450 nm in a microplate reader. The total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date.

Quoted performance characteristics include a sensitivity of <0.103 ng/ml and an assay range of 0.312–20 ng/ml. Reported sample recovery ranges (n=5) are: Serum 80%–102% (average 91%); EDTA Plasma 81%–100% (average 90%); Heparin Plasma 80%–89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was reported as: Serum 87–91%, 87–107%, 74–101%, 92–97%; EDTA Plasma 90–105%, 84–101%, 90–101%, 79–108%; Heparin Plasma 84–95%, 92–105%, 82–105%, 89–91%.
