Sandwich ELISA for the in vitro quantitative determination of human Histidine Decarboxylase in serum, plasma or other biological fluids, supplied by Antibodies.com. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for Histidine Decarboxylase pre-coated onto a microplate. Standards and test samples are added into the wells and any Histidine Decarboxylase present is bound by the immobilized antibody. After washing, a Biotinylated Anti-Histidine Decarboxylase Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Histidine Decarboxylase captured in each well, and concentration is calculated from the O.D. absorbance at 450 nm in a microplate reader using a standard curve.

Key specifications include an assay range of 2.34–150 ng/ml, a sensitivity of <0.7 ng/ml, and a total assay time of 4h 30m. The platform is a pre-coated microplate arranged as 12 x 8 well strips, read on a colorimetric microplate reader. Recovery data is provided across sample types: serum (80%–102%, average 91%, n=5), EDTA plasma (81%–100%, average 90%, n=5) and heparin plasma (80%–89%, average 84%, n=5). Dilution linearity is reported for serum, EDTA plasma and heparin plasma samples at 1:2, 1:4, 1:8 and 1:16 dilutions.

Components include a pre-coated 96 well microplate, lyophilized standard, detection solution A, detection solution B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers. Storage conditions for kit components are +4°C or –20°C as instructed.
