Human Histidine Ammonia Lyase ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human Histidine Ammonia Lyase (also referred to as Histidase) in tissue homogenates, cell lysates, or other biological fluids, with reactivity confirmed for human samples.

The assay employs a pre-coated 96-well microplate with an antibody specific for Histidase. Standards and test samples are added to the wells, where the target protein is captured by the immobilized antibody. After washing, a Biotinylated Anti-Histidase Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Histidase captured in each well. Absorbance is read at 450 nm in a colorimetric microplate reader, and concentration is calculated by reference to a standard curve. The complete assay time is 4h 30m.

Manufacturer-reported performance characteristics include a sensitivity of <0.62 ng/ml and an assay range of 1.56–100 ng/ml. Spike-and-recovery testing (n=5) showed average recovery values of 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%), and 84% in heparin plasma (range 80–89%). Dilution linearity across 1:2 to 1:16 dilutions was demonstrated for serum, EDTA plasma, and heparin plasma (n=5). Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
