From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human High Density Lipoprotein (HDL) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The kit uses a pre-coated 96-well microplate (12 x 8 well strips) and a biotinylated anti-HDL antibody paired with HRP-avidin detection. Colour development relies on TMB substrate, with the enzymatic reaction read as O.D. absorbance at 450nm in a microplate reader, and HDL concentration calculated by reference to a standard curve.

Reported performance figures include a product range of 93.8-6000 ng/ml and a sensitivity of 42.35 ng/ml, with a total assay time of 4h 30m. Components should be stored at +4°C or -20°C as instructed. Provided kit components include a pre-coated microplate (12 x 8 well strips), lyophilized standard (2 vials), Detection Solution A (120µl), Detection Solution B (120µl), 30X Wash Buffer (20ml), Sample Dilution Buffer (45ml), TMB Substrate (9ml), Stop Solution (6ml), and 5 adhesive plate sealers. Average spike-and-recovery across serum (n=5), EDTA plasma (n=5) and heparin plasma (n=5) ranged from 84% to 91%, and dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 in all three sample types.
