Human Hexokinase 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Hexokinase II in serum, plasma, tissue homogenates, or other biological fluids. The kit employs a pre-coated 96-well microplate to which standards and samples are added; Hexokinase II present in each sample is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-Hexokinase II Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate is then added to visualise the enzymatic reaction, producing a blue colour that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the Hexokinase II concentration is calculated by reference to a standard curve.

The assay has a sensitivity of <12.9 pg/ml and a measuring range of 31.2-2000 pg/ml. Total assay time is 4 hours 30 minutes. Detection is colourmetric and the kit is for use with human samples. Kit components are stored at +4°C or -20°C as instructed; the kit should not be used past its expiration date.

Spiked recovery was assessed in serum (n=5), EDTA plasma (n=5), and heparin plasma (n=5), with average recoveries of 91%, 90%, and 84% respectively. Linearity of dilution was evaluated at 1:2, 1:4, 1:8, and 1:16 for the same sample types.
