The assay employs a sandwich enzyme immunoassay technique, in which the target is captured on a pre-coated 96-well microtiter plate, then detected using a biotinylated anti-Met/c-Met antibody and an HRP-avidin conjugate.

The enzymatic reaction is visualised with TMB substrate, producing a blue colour that turns yellow upon addition of stop solution; absorbance is read at 450 nm. The quantitative detection range is 15.6–1000 pg/ml, with a sensitivity of 5.8 pg/ml, read on a colorimetric microplate reader. Total assay time is 4h 30m, and kit components are stored at +4°C or -20°C as instructed.

Reported recovery values include Serum (80–102%, average 91%), EDTA Plasma (81–100%, average 90%) and Heparin Plasma (80–89%, average 84%) across 5 samples each. Linearity is reported across dilutions of 1:2, 1:4, 1:8 and 1:16 for these sample types.
