Human Hepatocyte Growth Factor ELISA Kit

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of human HGF in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs a sandwich enzyme immunoassay technique: an antibody specific for HGF is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where HGF present in each sample is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-HGF Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of HGF captured. The concentration of HGF is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Sensitivity is <12.6 pg/ml with a product range of 31.2-2000 pg/ml. In spike-and-recovery testing, average recovery was 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%) and 84% in heparin plasma (range 80-89%), across n=5 samples per matrix. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions. The total assay time is approximately 4h 30m and detection is colourimetric. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
